Knowing the stoichiometry of a macromolecular interaction is crucial to mechanistic understanding. A robust way to determine stoichiometry is multisignal sedimentation velocity AUC (MSSV). This workshop will cover the MSSV methods implemented in SEDPHAT. By monitoring multiple wavelengths or signals during an SV experiment, a global analysis can be performed that decomposes the usual c(s) distribution into component distributions that can be integrated to determine stoichiometries. The theoretical background of the method will be discussed along with experimental strategies and practical examples. Participants wishing to participate in demonstrations should expect to install the software and data on a Windows laptop.
